The film follows four interrelated stories that show the impact of AIDS on different segments of Indian society. The first story is about Birju (Shiney Ahuja), a migrant worker from rural Bihar who works as a construction laborer in Mumbai. He has a wife, Yamuna (Raima Sen), and a son back home, whom he visits once a year. He also has a secret affair with Divya (Sameera Reddy), a married woman from an affluent family. When Birju falls ill, he discovers that he has contracted HIV from Divya, who got it from her husband, Abhay (Irrfan Khan), a successful businessman who travels frequently and has multiple sexual partners. Birju decides to return to his village and reunite with his family, unaware that he is putting them at risk of infection.

The second story is about Abhay (Irrfan Khan), who is diagnosed with HIV after donating blood for his friend's surgery. He is shocked and ashamed by his condition, and tries to hide it from his wife, Divya (Sameera Reddy), and his colleagues. He also faces discrimination and stigma from the society and the medical system. He meets Imran (Arjun Mathur), a young activist who works for an NGO that provides counseling and support for people living with HIV/AIDS. Imran helps Abhay cope with his situation and encourages him to disclose his status to his wife and seek treatment.

The third story is about Divya (Sameera Reddy), who learns about her husband's infection from Imran (Arjun Mathur). She is devastated and angry, and confronts Abhay (Irrfan Khan) about his infidelity and betrayal. She also realizes that she has been unfaithful to him with Birju (Shiney Ahuja), whom she met at a dance class. She decides to get tested for HIV, and finds out that she is positive. She feels guilty and remorseful, and wonders how to deal with her situation.

The fourth story is about Mr. Nanda (Satish Kaushik) and Mrs. Nanda (Tanvi Azmi), an elderly couple who live in a posh apartment in Mumbai. They are proud parents of their son, who is studying abroad. They are also fond of their maid, Laxmi, who has been working for them for many years. One day, they receive a letter from their son, informing them that he is gay and that he has HIV. They are shocked and confused by this news, and do not know how to react. They also discover that Laxmi has HIV too, which she got from her husband, who died of AIDS. They are faced with a dilemma of whether to keep Laxmi or fire her.

The film ends with a message that AIDS does not discriminate between rich and poor, urban and rural, male and female, or gay and straight. It urges the viewers to be aware of the facts about HIV/AIDS, to practice safe sex, to get tested regularly, and to support those who are affected by the disease.
